Once in Minnesota, one of the best ways to get around is by car unless you will be remaining in the cities. If you plan to visit the lake areas then it's probably best to hire a car and drive there. There is a good network of roads in Minneapolis and most are well marked. Most vacation destinations are within 150 mile radius of the Twin Cities so driving distances are not far.
